Purpose and Background
There is convincing clinical evidence regarding the effects of incretin-mimetics on weight loss. It appears that the primary mechanism of action of single GLP-1RAs and dual glucose-dependent insulinotropic polypeptide (GIP)/GLP-1RAs is reduction in energy intake, while dual and triple combinations that include glucagon receptor agonists not only reduce energy intake but also increase energy expenditure. Findings from sufficiently powered clinical trials showed significant beneficial effects of incretin-mimetics on clinical aging-related outcomes, while data from earlier trials on the effects of these drugs on weight loss showed positive effects on the risk factors for diseases and conditions associated with aging. Several modern (GIP)/GLP-1RAs have been recently approved by the FDA for different indications and may become available to NIA-supported investigators in near future.
This workshop will discuss existing evidence in the topic area and ensuing discussions will inform NIA’s research directions on the effects of incretin-mimetics on aging-related mechanisms and disease and conditions associated with aging. Another important research topic is the potential differences in efficacy and safety of this class of drugs in older adults.
